Engineering a temperature-sensitive self-cleaving intein

Temperature sensitive (Ts) mutants are important tools used for elucidating the roles of proteins in their native environment. However, the difficulty in engineering and selecting Ts mutants has greatly limited their use. Our work has focused on generating and identifying Ts mutants of the engineered self-cleaving recA mini intein from Mtu. Here, we present two strategies for engineering Ts mutant inteins that can be used to target a broad range of heterologous proteins.
