Glycomics Data Analysis Tool (GDAT): A MATLAB based web application for glycomics data analysis

The glycome constitutes the entire complement of glycans and glycoconjugates that compose a cell, tissue or organism. With the advent of new technologies a large amount of data related to the glycome are being collected at the transcriptome level using RNA-seq and gene microarrays, and structure level using mass spectrometry and glycan/lectin microarrays. These data are being stored in various online repositories/databases. The relation between data collected using different modalities is unknown since there is no streamlined computational method to integrate knowledge from these different experimental techniques. With the goal of providing such computational tools for large-scale data analysis, this project begins the development of freely-available web based tools for glycomics data analysis. The focus is on data analysis tools for mass spectrometry (MS) analysis. This program can either perform a comprehensive analysis of a single MS spectra or it can be used to analyze multiple MS spectra. It uses either built-in glycan database using network generation algorithms or user defined glycan database constructed using a ‘building-block’ approach. Given an MS spectra input, the program rapidly identifies and quantifies glycans in the spectrum. These data are then analyzed based on specific sub-structures or residues that compose the glycan. The program thus allows the quantitation of the relative abundance of different groups of glycans from multiple MS data, thus providing a useful set of tools to determine the role of various glycan structures in cellular functions like adhesion, signaling and recognition.
